刘默芳,博士,现任中科院上海生物化学与细胞生物学研究所研究组长(PI)、研究员、博士生导师。2000年毕业于中科院上海生化所生物化学与分子生物学专业获得博士学位。2000年至2006年分别在美国国家健康研究院国立癌症研究所(NIH/NCI)、约翰霍浦金斯医学院从事博士后研究、研究助理工作。2006年回上海生化与细胞所工作,主要从事生物医学基础和应用研究,围绕男性不育与肿瘤发生等开展系统性探索,获得了前沿性进展,发表论文70余篇,包括以通讯/共通讯作者在国际知名学术期刊CellMol CellDev CellNat Cell BiolCell Res等发表论文30余篇,多次受邀重要国际学术会议大会报告、组织重要国际学术会议。

作为首席科学家承担了科技部重点研发计划重点专项项目上海市科委重大基础研究项目科技部重点领域创新团队等,作为项目负责人或课题组长承担了科技部、国家自然科学基金委、中国科学院及上海市科委科研项目20多项;先后入选国家杰出青年科学基金上海市优秀学术带头人国家百千万人才工程、国家万人计划科技创新领军人才等,获中科院优秀研究生指导教师上海市巾帼创新奖中国生命科学十大进展谈家桢生命科学创新奖全国妇幼健康科学技术奖自然科学一等奖等荣誉,担任JBCBOR等学术期刊编委、中国生物化学与分子生物学会理事会常务理事及中国RNA专业委员会主任。

报告题目/Title:

The new functions of RNA regulation in male germ cells and human male infertility

讲座摘要/Abstract:

Spermatogenesis is the most complex cell differentiation process in mammals, including mitosis, meiosis, spermiogenesis, sperm capacitation and activation. These sequential male germline development is programmed by a series of highly orchestrated regulatory events in gene expression at each developmental step, in which RNA regulation plays an indispensable role and essential for the production of healthy sperm. On the one hand, uncoupling between transcription and translation, is a unique feature of gene regulation during spermatogenesis. Due to DNA recombination in meiotic phase and chromatin compaction in post-meiotic phase, transcription becomes inhibited and thus such developmental steps highly relies on gene products that are transcribed earlier and stored in translationally inert messenger ribonucleoproteins (mRNPs) until needed for translation;  On the other hand, as a new class of gene regulators, small non-coding RNAs such as piRNAs and miRNAs, piRNA-binding protein Piwi, and other RNA-binding proteins, are spatio-temporally expressed in mammalian male germ cells and play critical roles in regulating gene expression during spermatogenesis.  In addition, male infertility has become a global health problem, with main causes as oligospermia, asthenia, teratospermia or unknown cause.  Increasing evidence shows that gene mutations in RNA regulatory pathways are correlated to male infertility.  In this report, I will briefly summarize the recent research progress of our group on RNA regulation in spermatogenesis and male infertility, focusing on the new function of PIWI/piRNA in mammalian spermatogenesis and the discovery of male infertility-related Piwi gene mutations.
